Tymora bases their innovative, non-antibody technologies on soluble multifunctionalized nanopolymers (dendrimers) that allow phosphorylation detection and sample preparation with high reproducibility, specificity and, sensitivity.

Protein phosphorylation plays a critical role in the regulation of many cellular functions. As a result, detection of protein phosphorylation is essential in further understanding an organism’s signaling pathways and preventing and treating cancer and other abnormalities.

pIMAGO for universal, non-antibody detection of protein phosphorylation

Tymora Analytical offers pIMAGO — a novel, non-antibody, highly effective, and safe product for unbiased universal detection of any phosphoprotein.

pIMAGO is based on water-soluble, globular nanopolymers (i.e., dendrimers) that are multi-functionalized with reactive groups (e.g., Ti(IV) ions) for the highly specific recognition of phosphate groups, and with ‘reporting’ groups (e.g., biotin, fluorophore) that facilitate their detection. Multiple functionalized groups per dendrimer ensure very strong binding to the phosphoproteins (resulting in high specificity) and significant signal amplification from many “reporting” groups (resulting in high sensitivity).

One-Step PolyMAC for homogeneous enrichment of phosphopeptides prior to mass spectrometry analysis

PolyMAC is based on a metal-ion functionalized soluble nanopolymer to chelate phosphopeptides in a homogeneous aqueous environment. Compared to current strategies based on solid phase micro- and nano-particles, PolyMAC demonstrates outstanding reproducibility, exceptional selectivity, fast chelation times, and high phosphopeptide recovery from complex mixtures.

Due to low stoichiometry of phosphorylation, phosphopeptide enrichment is a critical step for successful phosphoproteomics experiments. Available here is a novel chemical strategy termed Polymer-based Metal-ion Affinity Capture (PolyMAC) for the isolation of phosphopeptides with exceptionally high reproducibility, selectivity, and sensitivity.
